چکیده
UNC119, also known as Retinal Protein 4 (HRG4) is significantly up-regulated in hepatocellular carcinoma (HCC) tissues. However, the clinical significance of UNC119 and its biological roles and associated mechanisms in HCC tumorigenesis remain unknown. In current study, quantitative real-time PCR, western bolt and immunohistochemical analyses were applied to evaluate the expression of UNC119 in HCC tissues and cell lines. The roles of UNC119 in cell proliferation, cell growth and cell cycle were analyzed by both loss- and gain-function assays in vitro and in vivo. Luciferase reporter assays and western blot were employed to investigate the mechanisms. We found that compared with normal liver, UNC119 is highly expressed in HCC tissues both in published GSE datasets and in our fresh or TMA tissues. We further found that highly expressed UNC119 is not only closely correlated with liver cirrhosis, tumor size and TNM stage but also predicts a poor prognosis of patients. Cellular function assays reveal that UNC119 promotes cell proliferation. In addition, it might through enhancing the activity of Wnt/β-catenin signaling to promote the progression of cell cycle and ultimate promotes cell proliferation and growth. Taken together, we found that UNC119 is commonly highly expressed HCC tissues and the expression pattern makes a difference in the prognosis of patients. Furthermore, it promotes cell proliferation and growth through Wnt/β-catenin signaling.

Angiopoietin-like protein 8 (betatrophin) may inhibit hepatocellular carcinoma through suppressing of the Wnt signaling pathway
Objective(s): Hepatocellular carcinoma (HCC) is one of the leading fatal neoplasms and the most common primary liver malignancy worldwide. Peptide hormone ANGPTL8 (betatrophin) may act as an important regulator in HCC development through the Wnt/β-catenin pathway. We aimed to evaluate the effects of recombinant ANGPTL8 on Wnt/β-catenin signaling in human liver carcinom...

Colorectal cancer (CRC) is one of the most common cancers, but the mechanisms underlying its initiation and progression are largely unknown. TGIF1 (TGFB induced factor homeobox 1) is a transcriptional corepressor that belongs to the three-amino acid loop extension (TALE) superclass of atypical homeodomains. It has been reported that TGIF1 is highly expressed in mammary cancer and non-small cell...

متن کاملSNF2H promotes hepatocellular carcinoma proliferation by activating the Wnt/β-catenin signaling pathway
Hepatocellular carcinoma (HCC) is one of the most common cancers worldwide and has an extremely poor prognosis. Surgical resection is always inapplicable to HCC patients diagnosed at an advanced tumor stage. The mechanisms underlying HCC cell proliferation remain obscure. In the present study, SWItch/sucrose nonfermentable catalytic subunit SNF2 (SNF2H) expression was tested in HCC tissues and ...
متن کاملDISC1 overexpression promotes non-small cell lung cancer cell proliferation
Neuropsychiatric disorder-associated disrupted-in-schizophrenia-1 (DISC1) activates Wnt/β-catenin signaling by inhibiting glycogen synthase kinase 3 beta (GSK3β) phosphorylation, and may promote neural progenitor cell and pancreatic β-cell proliferation. The present study found that DISC1 promotes non-small cell lung cancer (NSCLC) cell growth. Western blotting and immunohistochemistry analyses...
